Expansion of Clinical Trials for Ivonescimab
Summit Therapeutics Inc. has announced a significant extension of its clinical research efforts with the introduction of the global Phase III trial named HARMONi-GI3. This study aims to assess the efficacy of the investigational bispecific antibody, ivonescimab, when combined with chemotherapy in patients suffering from unresectable metastatic colorectal cancer (CRC). This initiative marks the fourth global Phase III study for ivonescimab and is particularly noteworthy as it ventures into colorectal cancer, an area where treatment options have been notably limited.
The HARMONi-GI3 trial will compare the combination of ivonescimab plus chemotherapy against the standard treatment of bevacizumab plus chemotherapy. The study is expected to enroll approximately 600 participants across multiple regions, with clinical trial sites set to activate in the United States by the end of the current year. The primary goal of this trial is to evaluate progression-free survival among participants.
Addressing a Critical Need in Cancer Treatment
Each year, around 48,000 individuals in the U.S. are diagnosed with unresectable recurrent metastatic microsatellite stable colorectal cancer (MSS CRC). This form of cancer has seen a dearth of approved therapies over the past two decades, particularly for first-line patients whose tumors lack specific biomarkers or activating mutations. Given this background, the introduction of ivonescimab presents a potential breakthrough, especially in light of the limited success of existing treatments like PD-1 inhibitors, which have not demonstrated significant benefits in this patient population.
At the recent 2024 Annual Congress of the European Society of Medical Oncology (ESMO), preliminary Phase II results were shared, highlighting the promising interaction of ivonescimab with FOLFOXIRI chemotherapy, yielding an objective response rate of 81.8% and a disease control rate of 100% among the initial cohort of 22 patients. These findings, which were collected exclusively by Akeso Inc., the company collaborating with Summit on ivonescimab, indicate a strong potential for this therapy within the colorectal cancer landscape.
